Unprecedented Powerful Characterization Method for Stem Cells (ES, iPS, MSC)

Glycan Profiling System

You may be not so familier with cell surface glycome, but a group of National Center for Child Health Development (NCCHD) and National Institute of Advanced Industrial Science and Technology (AIST) has already reported the potential in ISSCR 2009, and the results were inherited by NEDO iPS Project.(in Japanese), bringing in a lot of new insights furthermore.

A recent paper witten by Dr. Saito and Prof. Asashima et al. must be the first one which uncovers the relationships between gene expression patterns and cell surface glycome profilings in iPS, and another paper written by Dr. Toyoda and Dr. Umezawa et al. explains the performance of LecChip in characterizing pluripotent and multipotent stem cells.

Checking the phenotypes in terms of cell surface glycome is easier and simpler than investigating transcriptome and epigenetics, bacause glycan is the face of cell which intervenes cell-to-cell communication.

The profiling of cell surface glycome is so easy. What you have to do is only Cy3 fluorescence labeling onto cell lysate (prefer cell membrane fraction to whole cell lysate to improve accuracy). Glycostation is a trademark of GPB's glycan profiling system using an evanescent-field fluorescence excitation profiler (GlycoStation™ Reader 1200) and lectin microarrays (LecChip™).
